What it is
Mazdutide is a peptide made in a lab. It was first built by Eli Lilly and is being developed in China by Innovent Biologics. It copies oxyntomodulin, a natural gut hormone that attaches to both the GLP-1 receptor and the glucagon receptor. Natural oxyntomodulin lasts about 12 minutes in the blood. Mazdutide has changes and a fatty acid chain that keep it in the body for about 5 to 6 days. Almost all trials were done in Chinese adults. Other names: IBI362, LY3305677.

What the studies found
1. Weight dropped up to 11.7 percent in 12 weeks at a high dose.
A 2022 phase 1b study gave 24 Chinese adults with overweight or obesity high doses of mazdutide. In the 9 mg group, weight dropped 11.7 percent in 12 weeks, compared to 1.8 percent on placebo. No serious side effects were reported. Nausea, vomiting, and lower appetite were the most common. The study was very small. Human data.
2. Weight dropped up to 11.3 percent in 24 weeks.
A 2023 phase 2 trial gave 248 Chinese adults with overweight or obesity mazdutide at one of three doses, or placebo. After 24 weeks, weight dropped 6.7, 10.4, and 11.3 percent as the dose went up. The placebo group lost 1.0 percent. Waist size, blood pressure, blood fats, and liver enzymes also improved. Human data.
3. Weight dropped 14.0 percent in 48 weeks in a phase 3 trial.
The GLORY-1 trial gave 610 Chinese adults with overweight or obesity mazdutide at 4 mg or 6 mg, or placebo. At 48 weeks, weight dropped 14.0 percent at 6 mg. The placebo group gained a little weight. 49.5 percent at 6 mg lost at least 15 percent of their weight, compared to 2.0 percent on placebo. Liver fat also dropped. It was the first drug acting on both GLP-1 and glucagon receptors to finish a phase 3 obesity trial. Human data.
How much research exists
Phase 1, phase 2, and phase 3 human trials, almost all in China. Results in other populations have not been published. No heart outcome trial has been published.
